The Molecular Biology Program provides CRHMR investigators with assistance in DNA sequencing, known as the identification of linkage order of the nucleotides of a given DNA fragment.

Services

The Molecular Biology Program provides plasmid and PCR product sequencing services.

Only the facility’s manager has access to the sequencer, and therefore receives the samples and in return provides a complete range of services:

  • Purification of DNA to remove dNTPs and primers for PCR products;
  • Assaying to ensure DNA quality and quantity for plasmids and PCR products;
  • Sequenching using the BigDye Terminator v3.1 Cycle Sequencing kit in a thermocycler;
  • Purification of products obtained by ethanol/sodium acetate precipitation and centrifugation;
  • Analysis of samples already diluted in formamid, using the ABI 3100 sequencer.

Equipment

  • ABI 3100 sequencer
  • Thermocycler
  • Intelligent dark box LAS-3000 (Fuji)
  • Phosphor Imager Storm 840
  • Real-Time PCR iCycler (Bio-Rad)
  • Real-Time PCR model 7500 (Applied Biosystems)
  • Agarosis gel system
  • Alpha Imager
  • Spectrophotometer
  • Centrifuge with supporting table

The Molecular Biology Program of the Maisonneuve-Rosemont Hospital is also equipped with an ABI 3130XL sequencer that can be used when needed, and is compatible with the ABI 3100 sequencer.
